Spontaneous reactivation of the inactive X chromosome in mouse embryonal carcinoma cells.
The mouse embryonal carcinoma cell line MC12 carries two X chromosomes, one of which replicates late in S phase and shares properties with the normal inactive X chromosome and, therefore, is considered to be inactivated. Since the hypoxanthine phosphoribosyl transferase (HPRT) gene on the active X chromosome is mutated (HPRT(NDASH;)), MC12 cells lack HPRT activity. After subjecting MC12 cells t...
متن کاملX chromosome inactivation during induced differentiation of a female mouse embryonal carcinoma cell line.
P10 is a line of embryonal carcinoma cells with a euploid female karyotype. By making use of variant alleles of X-linked genes and of cytogenetic procedures, we have previously shown that the P10 cells have two genetically active X chromosomes. We show here that P10 cells rapidly differentiate into a cell type resembling extraembryonic endoderm when cultured in the presence of retinoic acid. Th...
متن کاملDehydroepiandroesteron increased proliferation of neural progenitor cells derived from p19 embryonal carcinoma stem cells.
Introduction: The p19 line of embryonal carcinoma cells develops into neurons, astroglia and fibroblasts after aggregation and exposure to retinoic acid (RA). Dehydroepiandroesteron (DHEA) is a neurosteroid, can increase proliferation of human neural stem cell (NSC) and positively regulated the number of neurons produced. This study was initiated to assess the effect of DHEA on neural progenito...
متن کاملReactivation of inactive X chromosome in buccal smear of carcinoma of breast
Buccal mucosal smears of 100 female patients of carcinoma of breast were compared with 100 controls matched accordingly. The frequency of Barr bodies was significantly lower in carcinoma of breast patients (menstruating and menopausal women) P < 0.001 when compared with controls indicating reactivation of the inactive X chromosome.
متن کاملLive imaging of X chromosome reactivation dynamics in early mouse development can discriminate naïve from primed pluripotent stem cells.
Pluripotent stem cells can be classified into two distinct states, naïve and primed, which show different degrees of potency. One difficulty in stem cell research is the inability to distinguish these states in live cells. Studies on female mice have shown that reactivation of inactive X chromosomes occurs in the naïve state, while one of the X chromosomes is inactivated in the primed state. Th...
